Overview

[18F]F-RESCA-RT4 (also referred to as [18F]AlF-RESCA-RT4) is a radiolabeled single-domain antibody (sdAb) tracer developed for immuno-PET/CT imaging of Trophoblast cell surface antigen 2 (Trop2), a pan-cancer biomarker implicated in tumorigenesis. The tracer is produced by conjugating the RT4 sdAb to a restrained complexing agent (RESCA), which allows efficient and mild radiolabeling with fluorine‑18 via an aluminum fluoride chelation method. This approach enables high radiochemical purity and stability, making it suitable for noninvasive visualization of Trop2 expression in vivo. Preclinical studies demonstrated specific tumor uptake in Trop2-positive models, with reduced renal accumulation due to the absence of a His-tag on RT4. The primary application is molecular imaging for cancer diagnosis and patient selection for Trop2-targeted therapies[1][2].
